Over the past 15 years, our laboratory as established and expanded on methodology to differentiate iPSC to GI tissues that include esophagus, stomach, small bowel and colon. Key advances establishing in vivo modeling in murine and rat host has introduced a functional ENS and established methods to provide a humanized immune response. The methods to isolate mesenchyme and endoderm provide a model to study key factors supporting engraftment and provides a potential product for both clinical and research studies. Recent methods to establish a robust tissue for engraftment utilizing a methods we term Confined Culture System provides both the ability to engraft all regionalized tissues and provide a functional ENS without the need to co culture neurospheres. Clinical translation of these models requires a GMP protocol and methods with a strategy to use both Allogenic and Autologous lines. We will present our ongoing efforts to achieve this.
