Oxidative damage to DNA: role of sequence and strand direction

ABSTRACT

Spontaneous oxidation of guanine residues in DNA generates 8-oxoguanine, a lesion related to carcinogenesis and aging. In this lecture, I presenta quantum mechanics/molecular mechanics (QM/MM) investigation of the oxidative damage caused by the reaction of water with ionized DNA. The results of the atomistic simulations elucidate the puzzling experimental evidence of an oxidative damage rate depending on the DNA sequence and strand direction.
