In specific aim 1, I propose the development of a miniaturized transient elastography (TE) probe, assembled using a small piezoelectric actuator. This probe can be attached to the tip of an intracardiac catheter for guiding cardiac therapies based on myocardium stiffness assessment. Preliminary results indicate that the small actuator is capable of generating measurable axial displacement that was used to distinguish between three tissue-mimicking phantoms with different elasticities and ex vivo porcine heart tissue under fresh and preserved conditions. In specific aim 2, I propose using shear wave elasticity imaging (SWEI) to monitor skin elasticity after auricular implantation, allowing for early assessment of skin damage and implant exposure. Preliminary mouse studies demonstrated the ability of SWEI to detect relative changes in skin elasticity before and after implantation of two constructs with different geometries. In specific aim 3, I propose to use SWEI to measure diaphragm elasticity after prosthetic repair in CDH patients.
