To understand immune regulation of cartilage regeneration, we propose the exploration of macrophage function in cartilage regeneration by: (1) developing murine osteochondral defect model with selective joint macrophage depletion to investigate the influence of macrophage depletion on autologous regeneration, (2) developing murine osteochondral defect model with macrophage-based therapy to investigate the influence of exogenous macrophage on cartilage regeneration, (3) investigating cartilage stem cells to explore possible interactions with macrophages.
