“Engineered Nanowires for miRNA-Mediated Control of T Cell Exhaustion” - Shaylyn Grier, Ph.D. student, Ankur Singh, Advisor.

Abstract

Persistent antigen exposure in cancer and chronic infection can drive T cell exhaustion, limiting the long-term effectiveness of adoptive cell therapies. A stem-like subset of exhausted T cells, known as progenitor exhausted T cells (Texprog), is associated with more durable immune responses and improved responses to immunotherapy, but is often present at low and variable levels. Here, we developed a silicon nanowire-based platform to deliver regulatory microRNAs directly into naïve mouse and human T cells, enabling early cellular programming before differentiation. Across mouse and human tumor models, including a three-dimensional diffuse large B-cell lymphoma organoid system, microRNA delivery increased features associated with Texprog, reduced signs of terminal exhaustion, improved cell survival, and maintained T cell function. These findings suggest that nanowire-mediated microRNA delivery can guide T cells toward a more persistent and therapeutically beneficial state, providing a promising strategy to enhance the effectiveness of adoptive T cell therapies.

"Transcriptional Drivers of Impaired B Cell Immunity in Lymphoma Survivors" - Rachel Ringquist, Ph.D., Ankur Singh, Advisor.

Abstract

R-CHOP, a combination of chemotherapy and the anti-CD20 antibody rituximab, is the standard first-line treatment for diffuse large B-cell lymphoma (DLBCL). While highly effective, recent studies have shown that patients treated with R-CHOP exhibit significantly diminished B cell responses even years after treatment concludes. Here, we aimed to identify transcriptional drivers of these diminished responses using an in vitro immune organoid platform that recapitulates B cell activation and maturation within the lymph node microenvironment. We profiled B cells from lymphoma survivors and found that our organoid system stratified donors into low, moderate, and high responders following influenza stimulation. Using single-cell RNA sequencing we identified unique transcriptional programs driving the observed functional differences between low and high responders. Furthermore, through single-cell BCR sequencing, we identified key differences in the clonal B cell responses between high and low responders. These results demonstrate that immune organoids can capture clinically meaningful heterogeneity in post-treatment B cell function, offering a platform for dissecting the mechanisms underlying impaired humoral immunity in lymphoma survivors.

This course module is designed for individuals interested in hands-on training in the fabrication of microfluidic devices using the soft lithography technique. This two day intensive short course will be structured to assume no prior knowledge of the technologies by the participants. The course agenda is evenly divided between laboratory hands-on sessions, including SU-8 master mold creation using photolithography and PDMS device fabrication in the IMS cleanroom, and supporting lectures. The goal for this course is to impart a basic understanding of soft lithography for microfluidic applications as practiced in academia and industry.  

Attendance is open to the general technical community and is not limited to current Georgia Tech students or IMS users. Anyone interested in soft lithography or microfluidics techniques is strongly encouraged to attend this course. The course is suitable for both new and experienced researchers interested in soft lithography techniques and applications. 

A course emphasis will be placed on IMS cleanroom resources, however, the concepts and techniques discussed are applicable to a broad array of research in this field.

Okorie "OkCello" Johnson is an Atlanta-based cellist, composer, looper, improviser, and storyteller who has been featured on the Tamron Hall Show, held a Kennedy Center Office Hours Artist Residency, presented at the Democratic Republic of Congo's 2022 Biennale, and co-composed and performed Liminal: An Atlanta Concerto with the Georgia Symphony Orchestra. He has scored an Emmy award-winning documentary, Imperfect Alibi; composed the original score for American Voices Against Apartheid, which premiered at the Apartheid Museum in Johannesburg before moving to the Kennedy Center; and scored a Suzi Bass Award-winning children's theatre production, Head to Toe for Alliance Theatres’ Theater for the Very Young.

His four studio albums — Liminal, Resolve, Beacon, and his most recent, Funny How Things Work Out (October 2025) — mark the milestones of an artist who has never stopped pushing. From concert halls to corporate keynotes, his cello has taken him everywhere. In 2024, that included Pixar in Oakland. In 2025, he headlined the Johns Creek Juneteenth Celebration, performed at the Aspen Institute Business Summit, and completed two concert voyages aboard the Ritz-Carlton Ilma superyacht. So far in 2026, he has taken his show to the Midwest, the Pacific Northwest, and Alaska — headlining performing arts centers in Rolla, MO, Eugene, OR, Fairbanks and Anchorage, AK to enthusiastic full houses.

He has opened for Maxwell, performed with India.Arie, De La Soul, and Big Boi of OutKast. Kate Kortum is a 23-year-old breakout jazz vocalist from Houston TX. She began her love for jazz and music at Houston's legendary High School for the Performing and Visual arts where she concentrated on her studies in flute and saxophone but ultimately fell in love with singing. Bringing Universal Diagnostics to the Point-of-Care: Raman Spectroscopy and Machine Learning

Raman spectroscopy of biofluids, cells, and tissues can diagnose a wide range of diseases – including diabetes, malaria, tuberculosis, celiac disease, cancer, and Alzheimer's – and quantify analytes such as glucose, albumin, and urea. This versatility has led to its description as a potential “universal” medical diagnostic. This thesis advances this vision along three fronts.

First, it introduces a method to improve the biological state resolution inferable by Raman imaging. While existing approaches commonly use spatial averaging to obtain a single spectrum, this work uses nonlinear dimensionality reduction to incorporate spatial information into the inferred cell state. Applied to imaging of egg maturation of a live C. elegans with BCARS – a highly sensitive and reproducible Raman imaging technique – this approach enables state resolution comparable to transcriptomics. This establishes the possibility that BCARS imaging combined with machine learning could serve as an omics surrogate in live cells.

Second, this machine learning approach is generalized into ScaleMAP, a dimensionality reduction technique that preserves local scale information typically erased by state-of-the-art methods such as UMAP, while retaining their neighborhood quality. Across diverse datasets, ScaleMAP recovers important structures that these techniques miss, including rare spectral “spikes” in Raman imaging, transitional "bridge" cells connecting distinct cell types in transcriptomics, and density structure spanning 17 orders of magnitude in flow cytometry.

Third, this thesis confronts the central barrier to real-world Raman diagnostics: the absence of an instrument that is simultaneously sensitive, reproducible, and affordable. The Raman diagnostics literature commonly uses instruments such as the Renishaw inVia Qontor, which can cost upwards of $200,000, yet exhibit limited reproducibility. This work demonstrates a proof-of-concept low-cost biofluid Raman spectrometer that surpasses the Qontor in both sensitivity and repeatability, at a current bill-of-materials of $1,300, with a roadmap to $150. It then leverages this repeatability to develop a proof-of-concept low-cost field calibration method for reproducibility across point-of-care centers.

Together, these contributions advance Raman diagnostics in two respects: the diagnostic information extractable from each measurement, and the translational path to obtaining measurements reliably in the real world.

The requirement for high levels of customer service, increasing numbers of SKUs and high labor costs have dramatically increased the complexity of warehouse operations. It is no longer sufficient to manage a warehouse based on a simple, arbitrary “ABC” classification of SKUs, which treats all those in a category as if they were identical. Instead, each decision – such as where to store or where to pick product – must be based on careful engineering and economic analysis. Each SKU must identify its own cheapest, fastest path through the warehouse to the customer and then compete with all the other SKUs for the necessary resources. Microfluidic-Enabled Adhesion Analysis of Therapeutic  T cells for Lymph Node Homing Potential

Adoptive cell therapies, including tumor-infiltrating lymphocytes (TILs) and chimeric antigen receptor (CAR)-T cells, have transformed cancer immunotherapy but remain limited by inefficient trafficking of transferred cells to therapeutically important tissues. In addition to direct tissue infiltration, effective T-cell therapies often depend on access to lymph nodes, which serve as specialized niches for immune surveillance, antigen presentation, T-cell maintenance and therapeutic activity across diverse disease contexts. Entry into lymph nodes is regulated by specialized vascular structures known as high endothelial venules (HEVs); however, current manufacturing approaches provide limited means to identify or enrich therapeutic T-cell populations with enhanced lymph node homing potential.

To address this gap, this dissertation develops microfluidic adhesion chromatography platforms that recreate vascular microenvironments under physiological flow to functionally assess T-cell trafficking potential and isolate cells based on adhesive behavior. Using expanded murine CD8⁺ T cells as a model of TIL products, we first demonstrate that adhesion to the HEV-associated adhesion molecule, nepmucin, identifies cell populations with enhanced early lymph node accumulation together with phenotypes associated with lymphoid cellular states. These findings were established using tumor-draining lymph nodes in a murine breast cancer model, providing a clinically relevant context for studying lymph node-directed trafficking during adoptive cell therapy. This framework is further extended to human CAR-T cells to compare adhesive phenotypes across lymph node- and tumor-mimicking vascular substrates. An integrated photoactivation strategy further distinguishes adhesive heterogeneity at both bulk-population and single-cell resolution.

Together, this work demonstrates that functional adhesion under physiological flow can distinguish T-cell populations with distinct tissue-trafficking behaviors that are not readily captured by conventional phenotypic analyses alone. By linking adhesive behavior with lymph node homing potential, this dissertation presents new tools for characterizing and enriching next-generation cellular immunotherapies with improved targeting of therapeutically relevant tissues.

Title: Self-assembly Across Scales: From Cytoskeletal Dynamics to Evolution of Multicellularity

Abstract: Philip Anderson famously remarked that "more is different". Complex systems exhibit emergent behaviors that cannot be understood solely from the properties of their individual parts. Living systems exemplify this principle, organizing molecules into cells and cells into multicellular structures through self-assembly. Yet the principles governing these emergent behaviors remain poorly understood. In this talk, I will describe our recent research, which combines quantitative biophysical experiments with theoretical modeling to uncover mechanisms underlying biological self-assembly across scales. At the molecular scale, I will discuss our work on the actin cytoskeleton, where we have shown that actin filament ends function as dynamic molecular ecosystems in which polymerases, depolymerases, and capping proteins collectively regulate actin filament dynamics. These findings challenge the classical model of actin treadmilling and reveal new mechanisms underlying cellular actin dynamics. I will then discuss our studies of the giant unicellular protist Stentor coeruleus, showing how hydrodynamic interactions between neighboring cells enhance feeding and generate asymmetric fitness advantages, providing a physical mechanism for the early transition to multicellular organization. Together, these studies illustrate the role of self-organization and collective interactions in driving the emergence of biological complexity across vastly different length scales.

Bio:  Dr. Shashank Shekhar is an Assistant Professor of Physics at Emory University. His research combines quantitative biophysical experiments with theory to uncover the physical principles governing biological self-assembly across molecular and organismal scales. His work has been recognized with the NSF CAREER Award, the NIH Maximizing Investigators' Research Award (MIRA), the Biophysical Society’s Early Career Award in Mechanobiology, the Whitman Early Career Award from the Marine Biological Laboratory and Les Grandes Avancées Françaises en Biologie award from the French Academy of Sciences. Dr. Shekhar received his Ph.D. in Nanobiophysics from the University of Twente (The Netherlands), following master's degrees in Nanoscience and Molecular Bioengineering from TU Delft (The Netherlands) and TU Dresden (Germany), and a bachelor's degree in Physics from Loyola College, India.

This workshop is intended to give C and Fortran programmers a hands-on introduction to MPI programming. Both days are compact, to accommodate multiple time zones, but packed with useful information and lab exercises. Attendees will leave with a working knowledge of how to write scalable codes using MPI – the standard programming tool of scalable parallel computing. It will have a hands-on component using the Bridges-2 computing platform at the Pittsburgh Supercomputing Center.

This event will be held in person at satellite sites, including Georgia Tech.

ABSTRACT
Deubiquitylases (DUBs) are a large family of enzymes that remove ubiquitin or ubiquitin chains from substrate proteins. Similar to other components of the UPS, DUBs have been associated with disease and pursued as drug targets. Despite enthusiasm around targeting DUBs both with inhibitors as well as DUBTACs, there remain unanswered fundamental questions around substrates and substrate scope as well as how DUBs work at a detailed mechanistic level. The overarching goal of our lab is to develop chemical tools that can be deployed to study basic function and translational potential of DUBs. Our platform integrates DUB library synthesis, medicinal chemistry, biochemistry, high-throughput screening, chemoproteomics, chemical genomics, structural biology, target validation and cancer biology. I will discuss recent discoveries enabled by our platform including new mechanism-of-action ligands for DUBs and their study in rare disease as well as new insights into DUB interplay with other components of the UPS.

BIO
Sara Buhrlage is an Associate Professor holding joint appointments in the Dana-Farber Cancer Institute department of Cancer Biology and the Harvard Medical School department of Biological Chemistry and Molecular Pharmacology. Buhrlage is a leader in harnessing deubiquitylases (DUBs) as emergent drug targets, where her lab has pioneered new approaches for interrogating DUB function, new insights into the chemical tractability of DUBs, and mapping the therapeutic potential of DUBs in cancer. In addition to research, she is dedicated to graduate education, holding leadership roles in multiple graduate programs and teaching graduate level courses. More recently, she has become involved in biotech and company creation as a scientific founder of Entact Bio. Buhrlage earned a Doctor of Philosophy in organic chemistry from the University of Michigan and completed a post-doctoral fellowship at the Broad Institute.

Title: Topological fault-tolerance in quantum computing

Abstract: An argument will be made that to build a utility scale quantum computer, topology is inevitable for error correction and fault-tolerance.  There are at least two ways to apply topology: via either non-abelian anyons in topological phases of mater or abelian anyons in traditional CSS quantum codes.  I will focus on the latter to show that geometrically enhanced topological quantum codes are good enough for current all-to-all platforms such as ion traps and neutral atoms, which liberate codes from the constraint of planar architecture and allow the exploration of higher dimensions. Therefore, building a useful quantum computer is now really an engineering problem of system architecture and control.

Bio: I am a professor of mathematics at UCSB, and former researcher of Microsoft Station Q. I am interested in quantum mathematics, and applying topology to quantum computing and physics.

Paper Craft Fundamentals is a hands-on workshop that introduces participants to essential techniques in paper-based art practices, including cutting, folding, scoring, rolling, weaving, and layering. Through guided demonstrations and self-directed experimentation, participants will develop a foundational understanding of these processes while exploring their potential for variation, combination, and personal expression.

The workshop is inspired by The Craft of Paper: Contemporary Takes on Tradition, an exhibition that examines how artists today are reinterpreting established paper craft techniques within contemporary art and design contexts. By the end of the workshop, they will have developed new technical skills as well as a deeper understanding of how paper-based practices continue to expand within contemporary creative fields.

 Please join us in celebrating The Craft of Paper: Contemporary Takes on Tradition, curated by paper artist Michael Velliquette.  The Craft of Paper explores modern interpretations of paper crafts, including cutting, folding, rolling, weaving, and layering.
