{"691943":{"#nid":"691943","#data":{"type":"event","title":"CANCELED - Coulter BME Seminar Series: Leslie Chan, Ph.D. ","body":[{"value":"\u003Ch2\u003EAbstract:\u003C\/h2\u003E\u003Cp\u003EThe gastrointestinal (GI) tract houses diverse intestinal enzymes from the human host and gut microbiome that play critical roles in mucosal immunity, host-microbiome interactions, drug metabolism, and digestion. Dysregulated intestinal enzyme activities give rise to a broad range of GI pathologies and can therefore serve as functional biomarkers of disease. However, current strategies for enzyme activity measurements often rely on invasive endoscopy to biopsy intestinal tissue samples for activity measurements in the clinical lab. As an alternative to endscopy, we have developed a new Volatile-releasing Activity-based Probe (VAPR) platform that enables remote sensing of pathology-causing intestinal enzyme activities via breath testing. The platform comprises ingestible sugar-based probes that sense and produce breath signals for specific intestinal glycosidase activities. In this talk, Leslie Chan will discuss the design and application of the probes for inflammatory bowel disease and for detecting harmful drug-metabolizing enzyme activity from the gut microbiome.\u003C\/p\u003E\u003Cp\u003E\u0026nbsp;\u003C\/p\u003E\u003Ch2\u003EBio:\u003C\/h2\u003E\u003Cp\u003ELeslie Chan\u2019s lab focuses on the development of bioresponsive nanomaterials that are delivered in vivo to harness biochemical cues in diseased tissue as triggers for disease readouts or site-specific therapeutic activity. They are especially interested in developing these tools to study, detect, and treat microbe-driven disease states (e.g. infections, microbiome dysbiosis, inflammation). Their goal is to develop solutions that can be readily translated at the point of care, for at-home use, or deployed for infectious disease surveillance to promote public health. Analog computing has shown great promise for low-power and energy-efficient computing, particularly in edge applications. However, large-scale demonstrations of these concepts remain limited due to the absence of a standardized design framework, unlike the well-established methodologies in digital computing. This work develops analog standard cells across various process nodes, extends analog synthesis tools to 16nm, and demonstrates several analog computing systems in both 16nm and 350nm process nodes. By utilizing analog standard cells and synthesis concepts, this framework enables designers without specialized analog circuit knowledge to build efficient analog computing systems. These devices are required to be compact and lightweight while also operating under extreme environmental conditions. Topology optimization, which has successfully been applied to the design of efficient lightweight structures, offers the potential to create heat exchangers with unconventional configurations that enhance overall system performance. This research explores the application of topology optimization to the design of high-performance heat exchangers. The proposed methodology is divided into three key stages. The first stage focuses on the development of a topology optimization framework based on a Darcy flow formulation for pipe configuration design of bi-fluid heat exchangers, exploring how variations in inlet location and pipe sizes could enhance thermal efficiency. The second stage integrates data-driven surrogates such as convolutional neural networks (CNN) with the topology optimization framework, improving the fidelity of the predicted flow fields. The final stage advances to a full-form design of a dual-flow 3D heat exchanger with arbitrary inlet conditions. This stage examines the additional efficiency gains through a combination of organic flow paths and flexible inlet configuration. The methodology utilizes both finite element and finite volume methods for the multiphysics analysis of the system, applying discrete and continuous adjoint formulations for sensitivity analysis respectively. The design objective is to optimize thermal efficiency by maximizing the average temperature and heat transfer rate under performance constraints, such as material strain energy distribution and power requirements. Computational analysis of menstrual blood loss and uterine vascular anatomy suggests that spiral arterioles are a plausible major source of menstrual bleeding. The high shear rates generated within these small vessels may also favor shear-induced platelet aggregation (SIPA), providing a potential mechanism for rapid hemostasis following endometrial shedding. An ongoing clinical study at Georgia Tech examines whether dysfunction in SIPA and primary hemostasis is present in a subset of women with HMB. Despite in vitro evidence of endogenous forces exerted by immune cells on immunoreceptors, in vivo measurement has been challenging due to technological limitations. Here, we present a novel technology platform based on synthetic Notch (SynNotch) circuits, enabling in vivo reporting of cell-generated forces on specific receptors. Our platform utilizes the mechano-sensitive properties of the Notch receptor, re-engineering it by replacing its ligand-binding domain with a single-chain variable fragment (scFv) of an antibody targeting the receptor of interest. The SynNotch receptor\u0027s activation induces the expression of a reporter gene. By adoptively transferring receiver cells expressing SynNotch into animals, we can measure the forces exerted by sender cells on the target receptor in vivo. We validated this platform using T cells as receiver cells and B cells as sender cells, with CD40 as the target receptor. Our results showed that B cells exert forces on CD40-SynNotch bonds, leading to the activation of SynNotch at forces around 2.4 pN per bond, increasing to a plateau at approximately 6.4 pN per bond. This mechanotransduction significantly influenced B cell signaling. Several significant difficulties complicate team members\u0027 ability to exchange information successfully. The DDC has come a long way since then, but the importance of sharing defensive driving principles with all drivers still has not changed.
