{"228951":{"#nid":"228951","#data":{"type":"event","title":"Micromechanical studies of protein-DNA interactions and chromosome structure","body":[{"value":"\u003Ch6\u003ESchool of Physics Colloquium Series: John Marko, Northwestern University\u003C\/h6\u003E\u003Cp\u003EThe centimeter-long DNAs in our cells are folded up into micron-scale chromosomes through an array of protein-DNA interactions. \u0026nbsp;Our group uses single-DNA micromanipulation \u2013 stretching and twisting of the double helix \u2013 as a tool to analyze a variety of enzymes acting on DNA. \u0026nbsp;I will describe a few different kinds of \u201cmagnetic tweezers\u201d experiments we are doing that are aimed at understanding enzymes that help to package DNA and to change its topology. We also use analogous but larger-scale micropipette-based micromanipulation approaches to study the large-scale structure of metaphase chromosomes.\u003C\/p\u003E\u003Cp\u003EI will discuss experiments that tell us the metaphase chromosomes behave as \u201cchromatin gels\u201d, apparently stabilized in part by DNA entanglement.
